I've now restored it to the C: drive and Starbound runs fine. I did notice, after all that, I had my documents, pictures and music folders set to a different drive other than the default C. After much tinkering about with folder & file permissions, trying EarthboundX's possible solution and even doing a reset of windows (for reasons other than trying to play Starbound) I still had the error.
